چکیده | ||
میکروارگانیسمهای نمکدوست و تحمل کنندهی نمک، در تطابق با تنش محیط، توانایی تولید مولکولهای فعال زیستی را دارند. همچنین نقش میکروارگانیسمها در زیستپالایی فلزات سنگین از جمله نیکل؛ که امروزه در مناطق شهری جزء آلودهکنندههای مهم آبهای سطحی و زیرسطحی به شمار میرود، شناخته شده است. هدف از این مطالعه، بررسی توانایی تولید مولکولهای فعال زیستی براساس تنوع ژن PKSدر میکروارگانیسمهای هالوتولرانت و هالوفیل مقاوم به فلز نیکل جدا شده از دریاچه نمک حاجعلیقلیخان بود. میزان عناصر آب و خاک دریاچه به وسیله دستگاه جذب اتمی اندازهگیری شد. میکروارگانیسمها از سه محیط کشت اختصاصی نمکدوستها MH ,SWNو MGMبه ترتیب با 10،2 و23 درصد نمک جدا شدند. سپس میکروارگانیسمهای مقاوم به عنصر نیکل غربال گردیدند. در مرحله آخر توانایی میکروارگانیسمهای مقاوم به نیکل در تولید متابولیتهای ثانویه با کمک آغازگرهای اختصاصی درون ژنی PKS مورد بررسی قرار گرفت. از میکروارگانیسمهای کشت شده، 50 جدایه از محیط کشتSWN ،22 جدایه از محیط کشت MH و 21 جدایه از محیط کشت MGM بدست آمد. آزمون مقاومت به فلز جدایهها نشان داد که 65%جدایه ها به فلز نیکل مقاوم بودند و بررسی مولکولی نشان دادکه برخی از این جدایهها توانایی تولید ترکیبات فعال زیستی را دارند. نتایج این تحقیق نشان میدهد که در دریاچه نمک حاجعلیقلیخان میکروارگانیسمهایی وجود دارندکه با توجه به مقاومت علیه فلز نیکل و توانایی در تولید محصولات پلیکتایدسنتازی میتوانند درزمینه زیستفناوری مورد استفاده قرار گیرند. | ||

Halophile and halotolerant microorganisms produce bio-active molecules under environmental stresses. The microorganisms play a role in bio-remediation of heavy metals including nickel; the most important water pollutant in urbans. Here studying of nickel resistant microorganisms from salt lake of Haj Ali Gholi Khan for their potential in bio-active molecules producing based on PKS genes variations is aimed. Chemical analyses using atomic absorption method for soil and water of the lake were done. The microorganisms isolated from MGM, MH and SWN culture media with 23, 10 and 2 % salt. The isolates were screened for nickel resistant’s followed by analyzing for presence of PKS genes using gene specific internal primers. Fifty, 22 and 21 microorganisms were isolated from SWN, MH and MGM medium respectively. 65% of the isolates were nickel resistant those carry the potential to synthesize bio-active compounds. These organisms because of their abilities in bio- remediation and also synthesizing bio molecules can be used in biotechnology. | ||
